Lysis in the NAO® Basket

NAO® Basket retains the lysis buffer during the lysis step of the forensic sample. Upon centrifugation on a high-speed centrifuge, valves in the bottom of the NAO® Basket open up and allow the sample lysate to pass through. The lysate containing the nucleic acids is efficiently recovered and collected in the provided collection tube, whereas the substrate remains in the NAO® Basket.

Improve DNA yield from challenging samples

The combination of NAO® Basket and FLOQSwabs® provides more than 90% DNA recovery of the reference sample collected by the swab, particularly critical for crime scene evidence testing. The 20 mm breaking point designed on the 4N6FLOQSwabs® is compatible with the NAO® Basket.


How to use NAO®Basket

1. Break the FLOQSwabs® at the breaking point into the NAO® Basket.
2. Add the lysis buffer and incubate as per extraction kit instructions.
3. Upon centrifugation, the sample lysate passes through the basket and is available for testing at the bottom of the tube.

Lyse & centrifuge in the basket

NAO® Basket are available in several formats as single pouches or bulk packages, both with 1,5 ml or 2 ml vials.



NAO® Basket is compatible with commercial Nucleic Extraction Platforms.

In addition, we developed the homemade protocol for DNA extraction as detailed below. Flocked swabs shafts were snapped at the molded breakpoint, inserted in the Nucleic Acid Optimizer NAO® basket (Copan), placed in microtubes and filled with 300 μL of saline solution, 300 μL of ATL buffer and 7 μL of Puregene Proteinase K 20 mg/mL (Qiagen).

Bloodstains and saliva stains collected with 4N6 FLOQSwabs® were extracted using Nucleic Acid Optimizer NAO® Baskets (Copan) and the QIAamp® DNA Investigator Kit. Modifications were made to the extraction protocol according to Copan’s recommendations. Modifications included performing sample lysis in NAO® Baskets under static conditions, with no vortexing or shaking during the first two incubation steps.

The manufacturer of the nylon flocked 4N6FLOQSwabs® advises the use of a Nucleic Acid Optimizer (NAO®) to increase the recovery of DNA from the swab. The NAO® is a semi-permeable basket that is applied in combination with high-speed centrifugation to collect the eluted DNA sample from the swab.
